2. Analysis Workflow (Total within 30 min)ProtocolThe Patho Gene-spin™ HS DNA/RNA Extraction Kit provides an easy and convenient protocol. For effective purification, add Proteinase K to biological samples, add Carrier RNA and Lysis Buffer, incubate at 56℃ for 10 minutes, and complete extraction through washing and elution steps.

Lysis & Binding Step
Add Proteinase K + sample (Blood/Tissue/Stool/Urine) + Carrier RNA/Lysis Buffer. Incubate at 56℃ for 10 min. Add Binding Buffer and load to spin column.

Washing Step
Wash sequentially with Washing Buffer A and Washing Buffer B to remove salts and PCR inhibitors. Centrifuge for column drying.

Elution Step
Elute with 40–50 µL of Elution Buffer. Store purified DNA/RNA eluate at -20℃ for immediate downstream diagnostics and research.

Performance Overview (Low-Titer Sensitivity & Flexible Elution)ValidationTo evaluate high-sensitivity extraction performance, low-titer pathogenic viruses (β-Coronavirus) and bacteria (C. chauvoei) were tested across vaccines, whole blood, and stool samples. The Patho Gene-spin™ HS kit demonstrated a more than 20% improvement in extraction sensitivity compared to previous compositions (sample count n=42; Previous 74% vs Patho Gene HS 95%).
A. High-Efficiency Viral RNA Extraction (β-Coronavirus)
Demonstrated significantly elevated extraction yield for low-titer β-coronavirus. Enhanced qPCR cut-off values (>2 Ct improvement) were observed in whole blood and stool matrices.
B. High-Efficiency Bacterial DNA Extraction (C. chauvoei)
Gram-positive bacteria (C. chauvoei) low-titer samples exhibited remarkable sensitivity improvements, especially in stool, showing an enhanced cut-off value (>3 Ct improvement).
Flexible Elution Volume Adjustment (40 µL vs. 50 µL)
Elution volume can be adjusted from 40 µL to 50 µL based on downstream requirements. While 50 µL is standard, reducing to 40 µL concentrates the purified nucleic acids to maximize detection sensitivity for trace-level samples.
✓ qPCR verification with pestivirus confirmed enhanced cut-off values and superior recovery rate (%) at 40 µL elution.
