Description

The LiliF® Blackleg Real-time PCR Kit is a diagnostic reagent designed for the qualitative detection of Clostridium chauvoei, the causative agent of blackleg, an acute and highly fatal bacterial disease in cattle. 

 

This assay specifically targets the cctA toxin gene of Clostridium chauvoei, enabling highly sensitive and specific detection of bacterial DNA. It is optimized for reliable diagnosis of blackleg in both beef cattle and dairy cattle, supporting rapid and accurate decision-making in veterinary diagnostics. 

 

This product is approved as a veterinary medical device (Approval No. 133-33).

1. Clinical Performance

 

•  Total samples : 408 clinical samples (308 positive and 100 negative, based on the reference method)

•  Clinical sensitivity : 100% (308/308, 95% CI : 98.62 ~ 100%)

2. Detection Performance

 

Stable Ct values were observed across different sample types and bacterial concentrations, demonstrating robust detection performance.

 


 

The assay demonstrated consistent detection performance across a wide range of clinical sample types, maintaining comparable Ct values regardless of sample type or target concentration. Reliable detection was achieved in various specimen types, including liver, heart, kidney, spleen, muscle, tissue homogenates, and cultured isolates derived from clinical samples. Furthermore, stable detection of Clostridium chauvoei was confirmed irrespective of cattle breed (beef cattle and dairy cattle), indicating robust performance across different animal sources.

TroubleShooting Guide
Q The target signal is positive, but the IPC (Cy5) shows a negative result. How should this be interpreted?
A - If a clear positive signal is observed in the FAM channel, the result can still be interpreted as positive. A negative IPC signal may occur due to signal suppression caused by strong target amplification or competition for PCR reagents during the reaction.

 

 

Q What do FAM and Cy5 signals represent in this assay?
A - The FAM channel is used for detection of Clostridium chauvoei (cctA gene). The Cy5 channel is used for the Internal Positive Control (IPC), which verifies proper PCR performance and detects potential inhibition.

 

 

Q What should I do if my instrument does not support the Cy5 channel?
A - The Cy5 channel is used for IPC detection. If your instrument does not support the Cy5 channel, IPC monitoring will not be available. It is strongly recommended to use an instrument compatible with both FAM and Cy5 channels to ensure reliable results.

 

 

Q What are the product’s shelf life and storage conditions?
A - The shelf life of this product is 12 months from the date of manufacture, and it should be stored frozen at –20 °C or below. When using frozen reagents, ensure that they are completely thawed to room temperature (15–25 °C) before use. Repeated freeze–thaw cycles may adversely affect product performance, so handle the reagents with care.
